The Mercedes-Benz R-Class (W251 standard wheelbase and V251 long wheelbase) was introduced at the 2005 New York International Auto Show. Positioned as a luxury 'Grand Sports Tourer', the R-Class blended the attributes of an MPV, a full-size station wagon, and a luxury SUV. It was offered in short and long wheelbase body variants with three-row seating configured for 6 or 7 passengers. Initial assembly took place at the Tuscaloosa plant in Alabama, USA.

Powertrains spanned 3.0L and 3.5L V6s, 5.0L and 5.5L V8s (R 500), and 3.0-liter V6 turbodiesels (R 280 CDI, R 320 CDI, R 350 CDI). Mercedes-AMG briefly offered the high-performance R 63 AMG equipped with a 510 hp 6.2-liter V8. A major 2010 facelift replaced the oval headlight styling with upright rectangular lighting and an assertive front grille. While European and US sales ended around 2012–2013, contract manufacturing for the Chinese market continued at AM General in Indiana until 2017.
