The first-generation Lexus HS 250h mid-size sedan (chassis code ANF10) unveiled at the North American International Auto Show in Detroit in January 2009 for the 2010 model year. It holds the distinction of being Lexus's first dedicated hybrid-only model built without a traditional internal combustion engine counterpart. Positioned between the IS and ES lineups on the Toyota New MC platform, its cabin utilized carbon-neutral plant-derived bioplastics for 30 percent of the interior surfaces alongside a pop-up navigation screen and Remote Touch controller.

Powertrain engineering combined a 2.4-liter 2AZ-FXE Atkinson-cycle four-cylinder gasoline engine with an electric motor to generate 187 total system horsepower delivered via an electronic CVT. A major 2013 mid-cycle refresh tailored for the Japanese domestic market integrated Lexus's spindle grille, LED headlight clusters, updated taillamps, and refined suspension dampers until model discontinuation in 2018.
